ABSTRACT

Abstract This study aimed to evaluate in vitro the plant growth promotion capacity of rhizospheric fungi from Opuntia cochenillifera. The isolates were submitted to phosphate solubilization, acetic acid-3-indole (AIA) production, antagonism against phytopathogens, and growth under water activity. All test results were subjected to statistical analysis. The studied isolates were able to solubilize phosphate, being F04 (Aspergillus sp.) and F05 (Coprinellus radians) the ones that presented the highest solubilization indices. As to produce AIA, F02 (Penicillium sp.) and F07 (Aspergillus sp.) were the most promising. All isolates could inhibit the growth of Colletotrichum sp. and Fusarium sp. Water restriction provided mycelial growth for all isolates. Therefore, these characteristics confer characteristics as growth promoters in plants.


Resumen El objetivo de este estudio fue evaluar in vitro la capacidad de los hongos rizosféricos aislados del cactus Opuntia cochenillifera para promover el crecimiento de las plantas. Los aislados fueron sometidos a pruebas de solubilización de fosfato, producción de ácido 3-indol acético (AIA), antagonismo contra fitopatógenos y crecimiento bajo actividad de agua. Todos los resultados de las pruebas se sometieron a análisis estadístico. Los aislados estudiados fueron capaces de solubilizar fosfato, siendo F04 (Aspergillus sp.) y F05 (Coprinellus radians) los que presentaron los mayores índices de solubilización. Sobre la producción de AIA, los que más produjeron fueron F02 (Penicillium sp.) y F07 (Aspergillus sp.). Todos los aislados pudieron inhibir el crecimiento de Colletotrichum sp. e Fusarium sp.. La restricción de agua proporcionó el crecimiento del micelio para todos los aislados. Por lo tanto, estes aspectos les confieren características como promotores del crecimiento en las plantas.

ABSTRACT

Among the factors that may influence the germination process of seeds, water deficiency is considered the most important. The present study aims to evaluate the physiological response of peanut genotypes to water deficiency. The experiment was conducted in a completely randomized design with four replications in a 12 x 7 factorial scheme with 12 peanut genotypes and seven polyethylene glycol 6000 concentrations corresponding to osmotic potentials of 0.00; -0.10; -0.20; -0.40; -0.60; -0.80 and -1.00 MPa. The data were submitted to analysis of variance and Tukey's test at a 5% probability level and, when significant, to analysis of variance adjusted to a quadratic polynomial curve. The germination percentage, seed vigor evaluated by the speed germination index, mean germination time, root length and root dry matter of some genotypes were affected by the osmotic potentials. Among the genotypes, there is a differential response related to the water deficiency level. In relation to the physiological response of the seeds, the lineages 506, 599, and 870 and the cultivar IAC Tatu-ST have a higher tolerance of water deficit. The peanut genotypes begin to suffer the effects of water deficiency at the osmotic potential of -0.4 MPa


Entre os fatores externos que podem influenciar o processo germinativo das sementes, a deficiência hídrica é considerada o mais importante. Dessa forma, o objetivo do presente trabalho foi avaliar a resposta fisiológica de sementes de genótipos de amendoim submetidos à deficiência hídrica. O experimento foi conduzido em esquema fatorial 12 x 7, tendo como tratamentos sementes de 12 genótipos de amendoim submetidos a sete concentrações de polietilenoglicol 6000, nos potenciais osmóticos correspondentes a 0,00; -0,10; -0,20; -0,40; -0,60; -0,80; -1,00 MPa. Constatou-se que os genótipos são afetados pelos potenciais osmóticos e, entre os genótipos, há resposta diferenciada em relação ao nível de deficiência hídrica. Em relação à resposta fisiológica das sementes, as linhagens 506, 599, 870 e a cultivar IAC Tatu-ST possuem maiortolerância à deficiência hídrica. Todos os genótipos de amendoim começam a sofrerem os efeitos da deficiência hídrica a partir do potencial osmótico de -0,4 MPa para a maior parte das características avaliadas

ABSTRACT

Primary polydipsia (PP) is marked by an increase in thirst, and most often presents in patients with psychiatric illnesses. Although uncommon in children, we experienced cases of PP in a 15-month-old boy and a 5-year-old girl. Both were admitted to the hospital with symptoms of polydipsia and polyuria that appeared 1–3 months before admission. Brain magnetic resonance imaging in both patients was normal. A water restriction test was performed after hospitalization and showed normal results. The symptoms improved after the parents were instructed to implement water-intake restriction for 2 weeks. Our report provides useful information for the treatment of PP in children.

ABSTRACT: The forest species Dimorphandra gardneriana Tul. is widely used for various pharmacological products, yet few basic studies have been undertaken to understand their ecological and physiological attributes under stress conditions. The goal of this research was to evaluate the seed germination and vigor when subjected to different osmotic potentials and temperatures. Water restriction was simulated with polyethylene glycol 6000 solution (PEG 6000) with osmotic potentials of 0; -0,2; -0,4; -0,6; -0,8, and -1,0MPa at temperatures of 20, 25, 30, and 35°C. The effect of the treatment was determined by the germination and vigor (germination speed index, length and phytomass of dry shoot and roots) of the seeds, in an entirely random design with four repetitions. From-0, 4MPa it occurs a drastic reduction in germination and vigor of seeds although these factors were less affected at temperature of 25°C up to -0, 2MPa.


RESUMO: A espécie florestal Dimorphandra gardneriana Tul. é bastante utilizada para diversos produtos farmacológicos, mesmo assim há poucos estudos básicos para compreender seus atributos ecológicos e fisiológicos sob condições de estresse. Desta forma, o objetivo nesta pesquisa foi avaliar a germinação e o vigor de suas sementes quando submetidas a diferentes potenciais osmóticos e temperaturas. A restrição hídrica foi simulada com solução de polietilenoglicol 6000 (PEG 6000) nos potenciais osmóticos de 0; -0,2; -0,4; -0,6; -0,8 e -1,0MPa nas temperaturas constantes de 20, 25, 30 e 35°C. Na avaliação do efeito dos tratamentos, determinou-se a germinação e o vigor (índice de velocidade de germinação, comprimento e fitomassa seca de parte aérea e raízes) das sementes, em delineamento inteiramente ao acaso, em quatro repetições. A partir de -0,4MPa ocorre redução drástica da germinação e vigor das sementes, fatores estes menos afetados na temperatura de 25°C até-0,2MPa.

ABSTRACT

The pathogenic fungi may be associated with the seeds during development, harvest and storage, and it can be disseminated through them. This work evaluated the seeds performance of three hybrid corn cultivars, BM 840 PRO, BM 810 and BM 207, submitted to different inoculum potentials of Fusarium verticillioides. Seeds were uniformly distributed over the medium fungal colony developed on PDA (potato dextrose agar) containing mannitol in water potential -2.0 MPa. Seeds remained in this way for different periods of contamination (0, 24, 48, 72 and 96 hours). Before and after seven months of seed storage the evaluations were performed. Seeds treated with fungicide Derosal Plus® (carbendazim + thiram, 250 mL p.c. 100 kg-1 of seeds) were used as control. The increased of inoculum potential of F. verticillioides fungus on corn seeds caused reduction in physiological quality in the potentials 72 and 96 hours of exposure before and after storage. The highest Fusarium verticillioides incidence is observed in non-stored seeds. Fusarium verticillioides incidence on corn seeds accentuates the deterioration in storage.


Os fungos fitopatogênicos podem associar-se as sementes durante o desenvolvimento, colheita e armazenamento, e por meio dessas serem disseminados. Este trabalho avaliou o desempenho de sementes de três cultivares de milho híbrido, BM 840 PRO, BM 810 e BM 207, submetidas a diferentes potenciais de inóculo de Fusarium verticillioides. As sementes foram distribuídas uniformemente sob colônia fúngica desenvolvida em meio BDA, contendo manitol no potencial hídrico -2,0 MPa. As sementes permaneceram desse modo por diferentes períodos de contaminação (0, 24, 48, 72 e 96 horas). Antes e após sete meses de armazenamento das sementes as avaliações foram realizadas. As sementes tratadas com o fungicida Derosal Plus® (carbendazim + tiram, 250 mL p.c. 100 kg-1 de sementes) foram usadas como testemunhas. O aumento do potencial de inóculo do fungo F. verticillioides nas sementes de milho causa redução da qualidade fisiológica nos potenciais de 72 e 96 horas de exposição, antes e após o armazenamento. A maior incidência de Fusarium verticillioides é observada nas sementes não armazenadas. A incidência de Fusarium verticillioides em sementes de milho acentua a deterioração no armazenamento.

Currently, the use of herbicides is essential in a practical and common in agricultural areas, but efficiency of these herbicides can be compromised when applied on plants that thrive in water deficit conditions, due to low uptake and translocation of the product. Therefore, the aim of this study was to compare the efficiency of control ACCase inhibiting herbicides applied post-emergence in plants of Eleusine indica under different soil water contents. The experiment was conducted in a greenhouse and the experimental design was completely randomized design with four replications, consisting of a 9x4 factorial, with the combination of three soil water potentials (-0.03, -0.07 and -1.5 MPa) three herbicides (fluazifop-p -butyl, haloxyfop-methyl and sethoxydim + oil) and four doses (0, 25, 50, and 100 % of the recommended dose). Herbicide application was made in plants in vegetative stage 2-3 tillers. The soil water potential was initiated in the development stage of two leaves, and the water was supplemented until the soil reaches the potential of - 0.01 MPa, when it came to minimum pre-determined for each water management. The physiological parameters evaluated were: photosynthetic rate, stomatal conductance, transpiration leaf temperature and plant dry mass. The visual assessments of phytotoxicity were performed at 7 and 14 days after application. The herbicides behaved in different ways according to the used water management. In severe water stress conditions (soil moisture at 8%) only fluazifop-p-butyl herbicide achieved satisfactory control (>90%) in E. indica plants.


O objetivo deste estudo foi relacionar a eficiência de controle de herbicidas inibidores da ACCase aplicados em pós-emergência em plantas de Eleusine indica submetidas a diferentes teores de água no solo. O experimento foi conduzido em casa de vegetação e o delineamento experimental utilizado foi o inteiramente casualizado, com 4 repetições, constituído de um fatorial 9X4, sendo a combinação de três manejos hídricos (-0,03; -0,07 e -1,5 MPa) com três herbicidas (fluazifop-p-butil, haloxyfop-methyl e sethoxydim + óleo mineral Assist) e quatro doses destes (100, 50, 25 e 0% da dose recomendada). Os manejos hídricos foram iniciados no estádio de desenvolvimento de duas folhas, repondo-se a água até o solo atingir o potencial de -0,01 MPa, quando este chegasse à tensão pré-determinada para cada manejo hídrico. Os parâmetros fisiológicos avaliados foram: taxa fotossintética, condutância estomática, transpiração, temperatura da folha e matéria seca das plantas. As avaliações visuais de fitotoxicidade foram realizadas aos 7 e 14 dias após a aplicação. Os herbicidas comportaram-se de maneiras distintas de acordo com o manejo hídrico utilizado. Em condições de estresse hídrico severo apenas o herbicida fluazifop-p-butyl atingiu controle satisfatório (>90%) em plantas de E. indica submetidas à estresse hídrico severo (8% de umidade no solo).

Este projeto objetivou relacionar a eficiência de controle de herbicidas inibidores da ACCase aplicados em pós-emergência em plantas de Cenchrus echinatus submetidas a diferentes teores de água no solo. Os experimentos foram conduzidos em casa de vegetação, com a aplicação de três diferentes herbicidas (fluazifop-p-butil, haloxyfop-methyl e sethoxydim + óleo mineral Assist). O delineamento experimental utilizado para cada herbicida foi inteiramente casualizado, com 4 repetições, constituído de um fatorial 3 x 4, sendo a combinação de três manejos hídricos (-0,03; -0,07 e -1,5 MPa) e quatro doses destes produtos (100, 50, 25 e 0% da dose recomendada). A aplicação dos herbicidas foi efetuada no estádio vegetativo de 2-3 perfilhos. Os manejos hídricos foram iniciados no estádio de desenvolvimento de duas folhas, repondo-se a água até o solo atingir o potencial de -0,01 MPa, quando este chegasse à tensão pré-determinada para cada manejo hídrico. As avaliações visuais de fitotoxicidade foram realizadas aos 7, 14, 21 e 28 dias após a aplicação e a matéria seca das plantas ao final destas. ABSTRACT

A deficiência hídrica do solo por ocasião do plantio e associação de patógenos com sementes podem ocasionar a diminuição da germinação e vigor de um lote de sementes, levando à redução do estande e introdução de doenças no campo. Nesse trabalho utilizou-se sementes de algodoeiro cultivar Deltapine Acala 90 (suscetível a ramulose) e linhagem IAC 2226 (resistente a ramulose) para avaliar o desempenho das sementes mediante condições de deficiência hídrica (manitol,-0,8MPa), e presença de Colletotrichum gossypii var. cephalosporioides. Após a submissão a dificiência hídrica em substrato agarizado na presença e ausência do fungo, pelos períodos de 0, 24, 36 72 e 108 horas, foram avaliados: emergência de plantas, índice de velocidade de emergência (IVE), peso de matéria seca de parte aérea, sanidade e índice de doença. Verificou-se que com o aumento do tempo de deficiência hídrica na presença e ausência do patógeno a emergência, vigor e matéria seca de parte aérea de ambos os genótipos diminuiu, sendo mais acentuado na presença do fungo. Observou-se aumento na incidência de C. gossypii var. cephalosporioides à medida que se aumentou o período de deficiência hídrica. O índice de doença na cultivar suscetível foi de aproximadamente 40% no maior período de restrição hídrica, enquanto no material resistente o índice manteve-se praticamente constante em 12% durante todo o período de restrição hídrica.


Water deficit soil at planting and association with seed pathogens can cause a decrease in germination and vigor of a seed, leading to a reduction of the stand and the introduction of diseases in the field. In this work we used seeds of cotton cultivar Deltapine Acala 90 (susceptible to ramulosis) and IAC 2226 strain (resistant to ramulosis) to evaluate the performance of seeds under conditions of water stress (manitol, -0.8 MPa) and presence of Colletotrichum gossypii var. cephalosporioides. After submitting to water stress in agar substrate in the presence and absence of the fungus, for periods of 0, 24, 36, 72 and 108 hours, were sown evaluated: plant emergence, speed of emergence index , dry weight of aerial part, health and disease index. It was found that with increasing time of water stress in the presence and absence of the pathogen emergence, vigor and dry weight of aerial part of both genotypes decreased, being more pronounced in the presence of the fungus. There was an increase in the incidence of C. gossypii var. cephalosporioides as it increased the amount of water restriction. The rate of disease in susceptible cultivar was approximately 40% higher in the period of water restriction, while the resistant strains index remained almost constant at 12% throughout the period of water restriction.

ABSTRACT

PURPOSE : Treatment of primary nocturnal enuresis (PNE) includs folk remedies and various treatments based on pathogenesis. We assessed the therapeutic effect of nocturnal water restriction as the primary treatment of PNE. MATERIALS AND METHODS : From October 1998 to June 1999, 41 children with PNE (>3 wet nights per week) who visited Ewha Womans University Mokdong hospital and who had good compliances to nocturnal water restriction for 2 months were included. Before and during nocturnal water restriction, daily fluid intake and urine volume were recorded for 2 days every 2 weeks. Responses to nocturnal water restriction were classified according to the decrease of wet nights as complete (>90%), partial (50-90%) and no (<50%) response. Predictors such as age, sex, daytime voiding dysfunction, fluid intake, urine volume, maximum urine volume per void and fasting urine osmolality were evaluated. RESULTS : The response rate to nocturnal water restriction for 2 month was 82.9% (34/41) [complete response : 39.0% (16/41), partial response : 43.9% (18/41)]. The response rate to nocturnal water restriction was significantly higher in monosymptomatic PNE than polysymptomatic PNE and more effective in PNE with er nocturnal fluid intake, nocturnal urine volume, and maximum urine volume than lower nocturnal fluid intake, nocturnal urine volume and maximum urine volume per void (p<0.05). Nocturnal urine volume, maximum urine volume per void and fasting urine osmolality after nocturnal water restriction has significantly increased higher in complete response and partial response group than in no response group (p<0.05). CONCLUSION : The nocturnal water restriction was effective in monosymptomatic PNE with nocturnal polydypsia, nocturnal polyuria and high bladder capacity.

ABSTRACT

BACKGROUND: Although the water restriction test(WRT) has been used as a standard test for the differential diagnosis of diabetes insipidus(DI), the measurement of plasma ADH concentration is also known to be useful method for differential diagnosis. Recent studies have shown that some patients with idiopathic central DI(CDI) were found to have a lesion on follow-up imaging studies. There have been no report in Korea on plasma ADH measurement for the differential diagnosis of DI, nor on follow-up imaging study of the idiopathic CDI. METHODS: We retrospectively reviewed the clinical and laboratory findings of 26 patients(12 men, 14 women, age 9-65 years) with CDI, including pituitary MRI or CT scan, who had been diagnosed with WRT and had undergone plasma ADH concentration measurement. RESULTS: 1) Clinical features of the patients with complete CDI did not differ from those of patients with partial CDI. 2) Maximal urine osmolality of complete CDI and partial CDI were 168+/-69mOsm/kg and 431+/-141mOsm/kg, respectively, and the percentage increase in the urinary osmolality after ADH injection was 209+/-149% and 29+/-17%, respectively. 3) Among the 26 patients, 10 patients had their plasma ADH measured. Nine patients in this group were diagnosed as CDI by WRT and plasma ADH concentration of the 9 was compatible for CDI. The plasma ADH level was also inappropriately low in one patient who had been diagnosed with primary polydipsia by WRT, the patient was diagnosed as partial CDI. 4) The findings of follow-up MRI revealed isolated thickening of the pituitary stalk in two cases of idiopathic CDI diagnosed initially with MRI. CONCLUSION: This study suggests that the measurement of plasma ADH can ensure a better differential diagnosis between partial CDI and primary polydipsia, and that the patients with idiopathic CDI should be examined regularly with MRI brain scan, including the pituitary gland.

ABSTRACT

Objective To investigate the effect of stress induced by water restriction on the level of phosphorylated(ERK1/2) in rat brain. Methods Fifty-eight male Wistar rats were used and randomly divided into control group(C,n=4) and three experimental groups: TW(drinking at scheduled time),EB(empty bottle stimulus),WR(restricted water drinking),which were subdivided into three subgroups by time as 3d,7d, 14d(n=6 for each).In C group,water was freely accessed to the animal for one week.In the experimental groups the water supply was restricted following a week of accommodation and the other week for timed water supply training.In TW group,the water was supplied twice a day and 10 min for each in a fixed time. In EB and WR groups,the water was served only once a day for 10 min,and in the other scheduled time for water supply only an empty bottle without water was provided in the EB group and water was limited in the WR group.All the animals were deeply anesthetized 1h after the last stimulus and transcardially infused,then the brains were cut into free-floating sections and processed for immunohistochemical staining of pERKI/2 with ABC method.The expression of pERK1/2 positive staining was measured with image analysis system. Results The area of phosphorylated ERK1/2 was significantly increased in many brain structures,especially in paraventricular hypothalamic nucleus(PVN),supraoptic nucleus(SON),ventrolateral septum(LSV),medial amygdaloid nucleus(MeA),central amygdaloid nucleus(CEA) and nucleus of solitary tract(NTS) in TW,EB and WR group compared with C group.The expression of phosphorylated ERK1/2 in SON,LSV, MeA and CeA in EB and WR group were higher than that in TW group;pERK1/2 expression of LSV,MeA,CeA and NTS in EB group increased and went down more early and quickly than that in WR group.The response of phosphorylated ERK1/2 was dependent on both stimulus methods and time in all the nuclei mentioned above except PVN,in which only the stimulus time was related. Conclusion The level of pERK 1/2 was significantly altered in PVN,SON,LSV,MeA,CeA and NTS following stimulation of water restriction.The results indicate that the activated nuclei in the brain by physical and psychological stressors are different.
